ISO 11290-1:2017/Amd. 1:2026 is an ISO amendment to the Part 1 detection method for Listeria monocytogenes and Listeria spp. in the food chain. It adds guidance on storing samples before analysis and updates the control strain used for performance testing of culture media and reagents. The document is aimed at laboratories and other users who apply the ISO 11290-1 detection method in food, feed, milk, surface and related microbiology work.

What does ISO 11290-1:2017/Amd. 1:2026 specify?

ISO 11290-1:2017/Amd. 1:2026 specifies changes to ISO 11290-1:2017, Part 1 of the horizontal method for detecting Listeria monocytogenes and Listeria spp. It updates the handling of samples before analysis and the quality assurance checks for media and reagents used in the method.

Clause or partWhat it covers
Clause 7Sampling, transportation and storage of samples
9.5.2.5.2 and 9.5.2.6Replacement of the control strain used in the method
Clause B.5Performance testing for culture media and reagents
Tables B.1 and B.2The detailed performance testing arrangements for enrichment, isolation and confirmation
BibliographyAdditional references for sampling, challenge tests and streak plating

The amendment keeps the focus on the detection method. It adds practical instructions for pre-analysis storage and aligns the quality-assurance strain set used during media and reagent testing.

What are the key requirements of ISO 11290-1:2017/Amd. 1:2026?

ISO 11290-1:2017/Amd. 1:2026 introduces a few practical points that matter directly in a test laboratory.

Sample handling before analysis

Clause 7 states that sampling itself is outside the method and should follow the relevant product standard, or an agreement between the parties concerned if no such standard exists. The clause also says the laboratory should receive a representative sample that has not been damaged or changed during transport or storage. In practice, this is the part that protects the validity of the result before the test even starts.

Clause 7 points users to ISO 7218 for storage conditions before analysis. If those conditions cannot be applied, other conditions may be used, but the laboratory must show that they do not change the contamination levels of Listeria spp. or Listeria monocytogenes, and the alternative conditions must be documented and justified in the test report.

Sampling references used with the method

Clause 7 names recommended sampling techniques in ISO/TS 17728 for food and animal feed, ISO 707 for milk and milk products, and ISO 18593 for surfaces. This matters because the method relies on a proper sample, but the sampling step itself is taken from the product-specific or surface-specific standard.

Control strain update

In 9.5.2.5.2 and 9.5.2.6, the amendment replaces Listeria monocytogenes WDCM 00109 with Listeria monocytogenes WDCM 00231. For users, this means the method now expects a different reference strain in the affected checks, so laboratory strain lists and media verification records need to follow the updated strain.

Performance testing of media and reagents

Clause B.5 says performance testing should follow ISO 11133, including its definitions of productivity, specificity and selectivity. Table B.1 covers the quality assurance of enrichment and isolation media, while Table B.2 covers media and reagents used in confirmation tests. In practice, this is the part laboratories use to verify that their media and reagents still work as intended.

Second isolation medium

Clause B.5 says the second isolation medium is tested according to the manufacturer’s specifications. This gives the laboratory a manufacturer-based route for the extra isolation medium rather than a single fixed testing pattern.

Extra strain option for Listeria spp.

A note in Clause B.5 says that, for productivity testing in the detection of Listeria spp., it can be informative to add another control strain from another Listeria species sensu stricto. This helps users check performance across the broader Listeria group when that is relevant to their work.

What terms does ISO 11290-1:2017/Amd. 1:2026 define?

  • Listeria monocytogenes - the target Listeria species named in the method.
  • Listeria spp. - the group of Listeria species covered by the method.
  • WDCM - the strain identifier used for reference culture collection strains in the tables.
  • Productivity - the ability of a medium to support growth as defined in ISO 11133.
  • Selectivity - the ability of a medium to inhibit unwanted organisms as defined in ISO 11133.
  • Specificity - the ability of the test to react as expected with the target organism, also used in ISO 11133 context.
  • Control strain - the reference strain used to check that media or reagents perform correctly.

Who uses ISO 11290-1:2017/Amd. 1:2026?

ISO 11290-1:2017/Amd. 1:2026 is used by microbiology laboratories testing food, feed, milk products and surfaces for Listeria. It is also relevant to quality managers who oversee media qualification and reagent checks, and to manufacturers or suppliers of culture media and test reagents who support the method.

The amendment is especially useful where laboratories need to decide how to store samples before analysis and how to document that storage, since both can affect whether Listeria is still detectable.

What changed in ISO 11290-1:2017/Amd. 1:2026 from the previous edition?

ISO 11290-1:2017/Amd. 1:2026 adds guidance for storage of samples before analysis in Clause 7. It also changes the control strain in 9.5.2.5.2 and 9.5.2.6 from Listeria monocytogenes WDCM 00109 to Listeria monocytogenes WDCM 00231.

Clause B.5 is rewritten to point users to ISO 11133 for performance testing of culture media and reagents, and the bibliography is expanded with references for sampling, challenge tests and streak-plate technique.

Which standards are used with ISO 11290-1:2017/Amd. 1:2026?

  • ISO 7218 - gives the storage conditions before analysis.
  • ISO/TS 17728 - gives sampling techniques for food and animal feed.
  • ISO 707 - gives sampling guidance for milk and milk products.
  • ISO 18593 - gives sampling techniques for surfaces.
  • ISO 11133 - gives the definitions and procedures for media and reagent performance testing.
  • ISO 20976-1 - is cited for shelf-life studies, which are outside the scope noted in Clause 7.

What does the ISO 11290-1:2017/Amd. 1:2026 document contain?

The amendment contains revised text for sample handling before analysis, including the link to ISO 7218 and the requirement to justify alternative storage conditions. It also contains updated strain references in the method and revised media and reagent performance testing text in Clause B.5.

The document includes Tables B.1 and B.2, which set out the testing approach for enrichment and isolation media and for confirmation-test media and reagents. Those tables support laboratory quality assurance, method verification and routine test system checks.

Replace the text with the following:
B.5  Performance testing for the quality assurance of the culture media and reagents
The definition of productivity, specificity and selectivity are specified in ISO 11133. In general, follow the
procedures for performance testing described in ISO 11133. Table B.1 provides the performance testing
for the quality assurance of the culture media for the enrichment and isolation. Table B.2 provides the
performance testing for the quality assurance of the culture media and reagents for the confirmation tests.
For the second isolation medium, use performance testing according to the manufacturer’s specifications.
NOTE For productivity testing, in the case of detection of Listeria spp., it can be informative to add another control
strain (from another Listeria species sensu stricto).
