Overview

SIST EN ISO 11348-2:2009/A1:2019 is an amended international standard developed by ISO and CEN focused on water quality analysis through a biological approach. Specifically, it outlines the method for determining the inhibitory effects of water samples on the bioluminescence of Vibrio fischeri bacteria using liquid-dried bacteria. This amendment updates the original ISO 11348-2:2007 standard to enhance precision and reliability in luminescent bacteria testing.

This method is widely recognized for environmental water monitoring and toxicity assessment, offering a sensitive and rapid bioassay to evaluate potential contamination and the presence of toxic substances compromising aquatic ecosystems.

Key Topics

  • Luminescent Bacteria Test: The procedure quantifies how water samples inhibit light emission from the marine bacterium Vibrio fischeri, a natural luminescent microorganism.

  • Use of Liquid-Dried Bacteria: The test utilizes liquid-dried bacteria preparations, which improve ease of handling, longer shelf life, and consistency compared to freshly cultured bacteria.

  • Inhibitory Effect Measurement: The focus is on determining the degree to which water contaminants reduce bacterial light emission - an indicator of toxicity or pollution in water samples.

  • Reproducibility and Accuracy Improvements: Amendment 1 refines criteria, such as limiting acceptable deviation in test sample dilutions to not exceed 3% points for reliable results.

  • Standardized Procedure for Water Quality Laboratories: The method includes detailed steps for sample preparation, incubation, measurement, and data interpretation to ensure consistent outcomes aligned with international water quality assessment protocols.

Applications

  • Environmental Water Monitoring: Applied by environmental agencies to assess the ecological safety of rivers, lakes, groundwater, and coastal waters.

  • Pollution and Toxicity Screening: Effective for detecting and quantifying the presence of toxic chemicals, industrial effluents, agricultural runoff, and other harmful contaminants.

  • Wastewater Treatment Evaluation: Used to monitor inhibitory effects of treated and untreated wastewater discharges on aquatic life.

  • Regulatory Compliance and Reporting: Supports compliance with environmental regulations by providing evidence-based toxicity data for water bodies.

  • Research and Development: Valuable tool in toxicological research, ecotoxicology studies, and water treatment technology optimization.

Related Standards

  • ISO 11348 Series: EN ISO 11348-2 is part of a series of standards specifying luminescent bacteria test methods for water quality; other parts cover different bacterial preparation techniques.

  • ISO/TC 147 Water Quality Standards: Provides a wider framework for biological, chemical, and physical water testing methods.

  • CEN/TC 230 Water Analysis Standards: European standards harmonizing water testing methods across countries to ensure consistent environmental monitoring.

  • ISO 5667 Series: Standards related to water sampling procedures complement the testing methodology by ensuring representative and uncontaminated samples.

  • ISO 10712: Guides the analysis of toxic substances in water, supporting comprehensive water quality assessment when paired with luminescent bacteria tests.

This amendment to EN ISO 11348-2:2008 ensures continued reliability and harmonization of the luminescent bacteria test using liquid-dried Vibrio fischeri, reinforcing its role as a critical biological method for detecting toxicity in aquatic environments. Understanding and applying this standard supports water quality professionals and laboratories in safeguarding ecosystem and public health through accurate toxicity evaluation.
