What are the key requirements of ISO 10519:2026?

ISO 10519:2026 is a wet-chemistry and spectrometric method built around a defined extraction and measurement sequence. In practice, the important points are sample preparation, fixed measurement wavelengths, and the calculation approach.

Sample preparation and sampling

Sampling is outside the method, but the laboratory sample has to be representative and protected from damage during transport and storage. ISO 664 is used to make the test sample from the laboratory sample after impurities are removed. If moisture exceeds a mass fraction of 10 %, the seeds are dried at 45 °C for 12 h to reduce the risk of chlorophyll loss.

Grinding and extraction

A 50 g portion of test sample is ground to a uniform seed meal. The test portion is then 2 g, weighed to the nearest 0.001 g, and extracted with 30 ml of extraction solvent. The method allows either a tube with steel balls shaken for 1 h or a commercial ball mill with steel balls for 20 min, so the user can choose the equipment that fits the laboratory.

Spectrometric measurement

The filtered extract is measured in an optical cell of at least 1 cm path length. Absorbance is read at 665 nm, with baseline correction using 705 nm and 625 nm. The spectrometer needs a suitable range from 600 nm to 700 nm and a spectral bandwidth of 2 nm, because the bandwidth affects the result.

Calculation and reporting

Chlorophyll content is expressed in mg/kg of product as received. The calculation uses the corrected absorbance, the cell path length, the test portion mass, the solvent volume, and a k factor given as 13 in the main method. If a result on a dry basis is needed, the moisture content is taken into account using ISO 665.

Precision and calibration

Annex A summarizes interlaboratory precision data for repeatability and reproducibility. Annex B gives a calibration-curve procedure for determining the k factor when the working spectrometer needs calibration. That matters when the instrument bandwidth or configuration means the default k factor is not suitable.

4 Principle
Extraction of a test portion in a suitable apparatus with a specified extraction solvent. Spectrometric
determination of the chlorophyll content of the extracted solution.
5 Reagent
Use only reagents of recognized analytical grade unless otherwise stated.

5.1 Extraction solvent.
Transfer to a 500 ml beaker 100 ml of anhydrous ethanol. Add to the contents of the beaker 300 ml of
anhydrous iso-octane (2,2,5-trimethylpentane) or anhydrous technical n-heptane or anhydrous petroleum
ether (essentially composed of C hydrocarbons, with a boiling range between 90 °C and 100 °C).
6 Apparatus
The usual laboratory apparatus and, in particular, the following shall be used.
6.1 Analytical balance, capable of weighing to the nearest 0,001 g.
6.2 Mechanical grinder, blade type, or coffee mill or equivalent.

Le présent document spécifie une méthode de spectrométrie permettant de déterminer la teneur en
chlorophylle des graines de colza. Elle n'est pas applicable à la détermination de la chlorophylle dans les
huiles.

4 Principe
Extraction d’une prise d’essai dans un appareil approprié, avec un solvant d'extraction spécifié.
Détermination spectrométrique de la teneur en chlorophylle de la solution extraite.
5 Réactif
Sauf indication contraire, utiliser uniquement des réactifs de qualité analytique reconnue.

5.1 Solvant d’extraction
Introduire 100 ml d'éthanol anhydre dans un bécher de 500 ml. Y ajouter 300 ml d'isooctane anhydre
(2,2,5-triméthylpentane) ou de n-heptane technique anhydre ou d'éther de pétrole anhydre (essentiellement
composé d'hydrocarbures C , ayant un intervalle d'ébullition compris entre 90 °C et 100 °C).
6 Appareillage
L’appareillage courant d’un laboratoire, notamment les éléments suivants doivent être utilisés.
6.1 Balance analytique, capable de peser à 0,001 g près.
6.2 Broyeur mécanique, à couteaux, ou moulin à café ou l'équivalent.
6.3 Micro-broyeur mécanique (voir Figure 1), composé de tubes en acier inoxydable ayant un volume
d'approximativement 50 ml et pouvant être bouchés hermétiquement, de billes pour roulements en acier
