Abstract

This document, part of EN 13130, specifies an analytical procedure for the determination of dimethylaminoethanol in food simulants: distilled water, 3 % w/v aqueous acetic acid, 15 % v/v aqueous ethanol and rectified olive oil. The method is appropriate for the quantitative determination of dimethylaminoethanol in an approximate analyte concentration range of 3,4 mg/kg to 36 mg per kilogram of food simulant.
NOTE   The method should also be applicable to other aqueous food simulants as well as to other fatty food simulants e.g. sunflower oil and a mixture of synthetic triglycerides.

SIST-TS CEN/TS 13130-19:2005 is a CEN Technical Specification published by SIST for measuring dimethylaminoethanol (DMAE) in food simulants used with plastics and articles in contact with foodstuffs. It specifies an analytical procedure that laboratories can use to determine DMAE in distilled water, 3 % w/v aqueous acetic acid, 15 % v/v aqueous ethanol and rectified olive oil, and to support migration testing for compliance work.

What does SIST-TS CEN/TS 13130-19:2005 specify?

SIST-TS CEN/TS 13130-19:2005 specifies a test method for the quantitative determination of DMAE in food simulants within an approximate concentration range of 3,4 mg/kg to 36 mg/kg of simulant. The method is written for migration testing from plastics, so the practical focus is on what has moved into the simulant after contact with a food-contact material.

The document is organized into the Scope, Normative references, Principle, Reagents, Apparatus, Samples, Procedure, Precision, Confirmation and Test report. It also carries a bibliography and should be read together with EN 13130-1.

What are the key requirements of SIST-TS CEN/TS 13130-19:2005?

  • Clause 3 uses solid phase extraction on a strong cation exchanger, then gas chromatography (GC) with flame ionization detection (FID) for measurement. In practice, the analyst must isolate DMAE from the simulant before GC analysis, and use an internal standard for quantification.
  • Clause 6 sets different sample preparation routes for aqueous simulants and olive oil. Aqueous samples are acidified and extracted directly; olive oil samples are first transferred into an aqueous phase before extraction. This matters because the same analyte needs different handling depending on whether the simulant is water-based or fatty.
  • Clause 6.4 requires calibration with analyte-free simulants spiked with known DMAE levels and the same internal standard. The two independently prepared DMAE stock solutions are used to generate two calibration series, which helps check that the calibration is reproducible.
  • Clause 7.1 and 7.2 require duplicate injections, stable GC conditions, and a rectilinear calibration graph with a correlation coefficient of 0,996 or better. For the reader, this means the result is not just one chromatogram point - the response must behave linearly across the working range.
  • Clause 7.3 allows result calculation either from a calibration graph or from regression parameters, with the regression approach preferred. The result is reported as DMAE in mg/kg of food simulant, and then converted to specific migration values when needed for comparison with the restriction criterion.
  • Clause 8 gives precision information from a collaborative trial and a within-laboratory detection limit range. This tells users what performance the method achieved in validation and helps quality managers judge whether their laboratory results are in a realistic range.
  • Clause 9 requires confirmation by GC-MS when the calculated specific migration exceeds 18 mg/kg. In practice, the GC-MS check is for identity and interference control, not for replacing the quantitative GC result.
  • Clause 10 defines the minimum test report content, including sample identity, analyte, method reference, results, and any confirmation or method modifications. This is what a laboratory needs for traceable reporting.

What terms does SIST-TS CEN/TS 13130-19:2005 define?

  • DMAE - Dimethylaminoethanol, the analyte measured in the food simulants.
  • DEAE - Diethylaminoethanol, used as the internal standard to correct the GC measurement.
  • GC - Gas chromatography, the separation technique used before detection.
  • FID - Flame ionization detector, the detector used for the routine GC measurement.
  • SIM - Selected ion mode, the GC-MS mode used for confirmation.
  • SML - Specific migration limit, the restriction value used when comparing the calculated migration result.
  • WDL - Within laboratory detection limit, the low-end capability reported in the precision section.

Who uses SIST-TS CEN/TS 13130-19:2005?

SIST-TS CEN/TS 13130-19:2005 is used by testing laboratories, food-contact material manufacturers, regulators, and quality teams working with plastics for food packaging and related articles. They use it to prepare migration samples, run GC measurements, confirm identity by GC-MS when needed, and report DMAE results for compliance evaluation.

Which standards are used with SIST-TS CEN/TS 13130-19:2005?

  • EN 13130-1:2004 - provides the general guide for test methods, exposure conditions, and conversion to specific migration values.
  • ISO 5725 - is used for the precision evaluation described in the performance section.
  • DIN 32645 - is used for the detection-limit estimation method cited in the precision section.

What does the SIST-TS CEN/TS 13130-19:2005 document contain?

The document contains reagent recipes, apparatus requirements, detailed sample preparation for aqueous simulants and olive oil, GC operating conditions, and calibration steps. It also includes example chromatographic conditions for two columns, a GC-MS confirmation approach with typical ions, and the formula-based calculation path for DMAE concentration. For laboratory use, it gives the reporting items needed to document the analysis and any confirmation work.

Introduction
Dimethylaminoethanol (DMAE), C H NO, PM/Ref. No 16150, is a monomer used in the manufacture of
4 11
certain plastics materials and articles intended to come into contact with foodstuffs. After manufacture
residual dimethylaminoethanol can remain in the finished product and may migrate into foodstuffs
coming into contact with that product.
The method has been pre-validated by collaborative trial with two laboratories using food simulants.
1 Scope
This document, part of EN 13130, specifies an analytical procedure for the determination of
dimethylaminoethanol in food simulants: distilled water, 3 % w/v aqueous acetic acid, 15 % v/v aqueous
ethanol and rectified olive oil. The method is appropriate for the quantitative determination of
dimethylaminoethanol in an approximate analyte concentration range of 3,4 mg/kg to 36 mg per
kilogram of food simulant.
NOTE The method should also be applicable to other aqueous food simulants as well as to other fatty food
simulants e.g. sunflower oil and a mixture of synthetic triglycerides.
2 Normative references
The following referenced documents are indispensable for the application of this document. For dated
references, only the edition cited applies. For undated references, the latest edition of the referenced
document (including any amendments) applies.

limitation – Part 1: Guide to test methods for the specific migration of substances from plastics to
foods and food simulants and the determination of substances in plastics and the selection of
conditions of exposure to food simulants.
3 Principle
Dimethylaminoethanol (DMAE) is extracted from the food simulants by means of solid phase extraction
on a strong cation exchanger. The absorbed DMAE is removed from the absorbent with triethylamine
and the resulting solution is submitted to gas chromatography to determine the amount of DMAE in the
simulants. For quantification purposes an internal standard is added. Calibration is achieved by analysis
of relevant simulants containing known amounts of DMAE.
Confirmation of dimethylaminoethanol is established by combined gas chromatography/mass
spectrometry.
