(Main)Soil improvers and growing media - Determination of chromium(VI)
- Abstract
This document specifies a method for the determination of hexavalent chromium (CrVI) in soil improvers and growing media using ion-exchange chromatography and subsequent UV-VIS detection. The method described is suitable to quantify the Cr(VI) content in soil improvers and growing media down to 1 mg/kg dry matter.
NOTE 1 Samples with a low dry matter content can have an influence on the achievable limit of quantification as defined in the scope.
The results obtained from this method are strictly dependent on the extraction conditions. Results obtained by using other extraction procedures (extraction solution, pH of the extraction solution, extraction time, extraction temperature, etc.) are not comparable with the results produced by the procedure described in this document.
NOTE 2 This method has been validated in an interlaboratory study with specific products that were present on the market during the study (as included in Annex C, biowaste compost dry HM spiked (Rottegrad 3, milled, sieved at 0,5 mm, dried and spiked), green compost dry HM spiked (Rottegrad 5, milled, sieved at 0,5 mm, dried and spiked), solid digestate dry (milled, sieved at 0,5 mm and dried), liquid digestate HM spiked (sieved 0 mm to 12 mm, spiked) and growing media blend 1 dry HM spiked (white peat, green compost, wood fibre, lime, inorganic fertilizer (NPK + micro elements), milled, sieved at 0,5 mm, dried and spiked)).

FprEN 18257 is a draft European Standard from CEN for determining hexavalent chromium (Cr(VI)) in soil improvers and growing media. It specifies an extraction followed by ion-exchange chromatography with UV-VIS detection, so laboratories can measure Cr(VI) on a dry-matter basis. The document is written for testing laboratories and for technical users who need comparable Cr(VI) results for these materials.
What does FprEN 18257 specify?
FprEN 18257 specifies a chemical method for measuring Cr(VI) in soil improvers and growing media. The method is designed to quantify Cr(VI) down to 1 mg/kg dry matter, but the achievable limit can be affected by samples with low dry matter content.
The standard also states that results depend strongly on the extraction conditions. That matters in practice because results from other extraction solutions, pH values, times or temperatures are not comparable with results obtained by the procedure in this document.
Sampling is handled by EN 12579, not by this method itself. Clause 7 points the user to that standard and emphasizes that the laboratory needs a representative sample that has not been changed during transport or storage.
| Annex | What it covers |
|---|---|
| Annex A | Chromatographic conditions for the direct detection method |
| Annex B | Chromatographic conditions for the method with post-column reaction |
| Annex C | Performance characteristics of the method |
What are the key requirements of FprEN 18257?
Extraction and sample handling
Clause 4 sets the principle: extractable Cr(VI) is leached with phosphate buffer at pH 7.0 to pH 8.0, then the filtered extract is analysed by ion-exchange chromatography with UV-VIS detection. The document also notes that a preliminary total chromium check in aqua regia extracts by ICP-AES, using prEN 13650:2025, can help decide whether full Cr(VI) testing is needed.
Clause 5 requires analytical-grade reagents and careful preparation of the extraction solution, standards and water. In practice, this means reagent quality, gas blanketing and pH control are part of the measurement result, not just laboratory housekeeping.
Clause 5 also specifies oxygen-free argon or nitrogen and prefers argon. That matters because the method is sensitive to reduction, so limiting oxygen and keeping extracts stable helps protect the Cr(VI) signal.
Apparatus and calibration
Clause 6 lists the equipment needed, including a shaker, pH meter, membrane filters, an ion-exchange chromatograph or HPLC with UV detector, and an analytical balance. A DAD is recommended, and that is useful because it helps confirm the chromate peak by its UV spectrum.
Clause 8.3 uses external standard calibration with at least five standards. The standards should cover the expected measurement range, and the same injection volume should be used consistently, which makes the calibration curve directly usable for the sample extracts.
Extraction and chromatography
Clause 8.1 gives different extraction steps for solid and shear-resistant pasty samples on the one hand, and liquid or aqueous pasty samples on the other. The key practical points are the sample mass, the extraction time, the immediate filtration, and the pH check after extraction; if the pH is outside 7.0 to 8.0, the procedure is repeated.
Clause 8.1 also says the instrumental determination should be carried out as soon as possible and within a day after extraction. That is important because Cr(VI) can be reduced after extraction, which would lower the reported result.
Clause 8.2 allows two tested chromatographic approaches: Annex A uses direct detection of the chromate peak at 372 nm, while Annex B uses a post-column reaction with 1,5-diphenylcarbazide and detection at 540 nm. For users, this means the standard accepts different laboratory instrument setups, but the method must still be verified by recovery testing.
Quality control and reporting
Clause 8.4 requires verification with a soluble Cr(VI) standard from a different source than the calibration standards. The QC sample must give a recovery between 80 % and 120 %, and matrix spikes are used routinely to check for sample-induced reduction or other matrix effects.
Clause 9 calculates the Cr(VI) content on a dry-matter basis using the sample moisture and dry matter content from prEN 13040-1:2025. The result is reported in mg/kg to the nearest 0.1 mg/kg, and the dry matter content is reported to the nearest 0.1 %.
Clause 10 defines what the test report must contain: the result, the standard reference, sample description, technique used, dry matter content, and any deviations or factors that could affect the result. If a sample shows highly reducing behaviour, that needs to be stated in the report because the native Cr(VI) content may not be quantifiable.
What terms does FprEN 18257 define?
- Chromium(VI) content - The amount of chromium(VI) in soil improvers and growing media after extraction with an aqueous salt solution at pH 7.0 to 8.0, reported as mg/kg dry mass.
- Cr(VI) - The hexavalent form of chromium that this standard measures.
- Dry matter - The mass basis used to express the result, so moisture must be measured or taken into account.
- Recovery rate - The percentage used to check whether the method is working correctly on a soluble Cr(VI) spike.
- Ion-exchange chromatography - The separation technique used to isolate chromate before detector readout.
- UV-VIS detection - The detector approach used to measure the chromate signal in the ultraviolet-visible range.
- DAD - Diode array detector, recommended in Annex A because it can help confirm the identity of the chromate peak from its spectrum.
- MWD - Multiple wavelength detector, used in Annex B with the post-column reaction method.
Who uses FprEN 18257?
FprEN 18257 is used by laboratories that test soil improvers and growing media for Cr(VI). It is relevant for routine quality control, product verification and compliance testing where a measurable and comparable Cr(VI) result is needed.
The method is also useful for manufacturers and technical buyers who need to understand how a Cr(VI) result was generated. Annex C shows validation on biowaste compost, green compost, solid digestate, liquid digestate and a growing media blend, so the standard is clearly aimed at these kinds of materials.
Which standards are used with FprEN 18257?
- CEN/TS 17732:2022 - Provides the terminology used for soil improvers and growing media.
- EN 12579:2024 - Covers sampling, which Clause 7 requires the user to follow.
- prEN 13040-1:2025 - Provides sample preparation and the dry matter and moisture values used in the calculation and expression of results.
- prEN 13650:2025 - Is named in Clause 4 for aqua regia digestion used in a preliminary total chromium check.
- ISO 5725-2 - Is used for the validation approach and the repeatability and reproducibility data in Clause 11 and Annex C.
- EN ISO 17075-2:2017 - Is cited as the source of the chromium(VI) content definition adapted for this document.
What does the FprEN 18257 document contain?
FprEN 18257 contains the analytical principle, reagent preparation, apparatus, sampling note, detailed extraction procedures, chromatography and calibration steps, recovery verification, calculation rules, reporting requirements and validation information. It separates the method into a general procedure and two annexes with example chromatographic settings.
Annex A gives a direct-detection chromatographic setup, including the detector wavelength and example operating conditions. Annex B gives a post-column reaction setup based on 1,5-diphenylcarbazide and shows the system layout used for that approach.
Annex C summarizes the interlaboratory validation, including the materials tested and the performance characteristics for repeatability and reproducibility. The annex also explains how the method behaved with different matrices and why matrix spikes are important when samples can reduce Cr(VI).

3 Terms and definitions
For the purposes of this document, the terms and definitions given in CEN/TS 17732:2022 and the
following apply.
3.1
chromium(VI) content
amount of chromium(VI) in soil improvers and growing media determined after extraction with an
aqueous salt solution at pH 7,0 to pH 8,0
Note 1 to entry: The chromium(VI) content is reported as chromium(VI) in milligrams per kilogram (mg/kg),
expressed as the dry mass of the sample.
[SOURCE: EN ISO 17075-2:2017, 3.1, modified: “leather” is replaced by “soil improvers and growing
media” and “pH 7,0 to 8,0” is replaced by “pH 7,0 to pH 8,0”]
4 Principle
Extractable Cr(VI) is leached from the sample in phosphate buffer at pH 7,0 to pH 8,0. An aliquot of the
filtered extract, or its dilution, is analysed for Cr(VI) using ion-exchange chromatography with UV-VIS
detection.
A preliminary determination of the total chromium in aqua regia extracts by ICP-AES (prEN 13650:2025)
can reduce the number of the samples where the determination of chromium(VI) is necessary because if
the content of aqua regia (total) extractable chromium is lower than the legislative limit for chromium(VI)
then the determination of chromium(VI) may be omitted.

with soil improvers and growing media. It is important that the laboratory receives a sample that is
representative of the product under consideration. The sample should not have been damaged or
changed during transport or storage.
8 Procedure
8.1 Preparation of analytical solution
8.1.1 Solid and shear resistant pasty samples
The operator shall weigh (6.9) approximately 10 g ± 0,1 g of fresh sample of soil improvers and growing
media to the nearest 0,01 g.
A higher sample intake may be considered for coarse samples to keep the liquid to solid ratio during the
extraction.
Following tasks shall be performed:
— Pipette 100 ml (V ) of degassed solution (5.1) into a 250 ml conical flask (6.2). If the sample is
expected to contain a significant amount of chromium(III), 1 ml of magnesium chloride solution (5.8)
may be added. Displace oxygen by passing oxygen-free argon (or nitrogen) (5.6) into the flask.
Remove the aeration tube (6.3), add the sample and close the flask with a stopper.
— Shake the conical flask with the sample for 30 min on a mechanical orbital shaker at
−1 −1
± 10 min (6.1) at room temperature to extract the Cr(VI).
100 min
— Shake the suspension in a smooth circular movement to keep the sample from adhering to the wall of
the flask and avoid shaking faster than specified.
Immediately after completing the 30 min of extraction, filter the contents of the conical flask through a
membrane filter into a glass or plastic vessel with lid. Check the pH of the solution. The pH of the
oSIST prEN 18257:2025
prEN 18257:2025 (E)
solution shall be between 7,0 and 8,0. If the pH of the solution is not within this range, start the
complete procedure again and consider using a smaller sample mass. If filtration is not possible in a
30 min period, centrifugation may be applied prior to filtration.
— Transfer an aliquot of the filtered extract, or its dilution, into a vial (6.11). Instrumental determination
of Cr(VI) should be performed as soon as possible and within a day after the extraction phase to avoid
risks of reduction.
8.1.2 Liquid and aqueous pasty samples
The operator shall weigh (6.9) approximately 10 g ± 0,1 g of sample of soil improvers and growing media
to the nearest 0,01 g.
The following tasks shall be performed:
— Pipette 100 ml (V0) of degassed solution (5.1) into a 250 ml conical flask (6.2).
— Shake the conical flask with the sample for 10 min ± 1 min on a mechanical orbital shaker at
−1 −1
100 min ± 10 min (6.1) at room temperature to extract the Cr(VI).
— Shake the suspension in a smooth circular movement to keep the sample from adhering to the wall of
the flask and avoid shaking faster than specified.
— Immediately after completing the 10 min of extraction, filter the contents of the conical flask through
a membrane filter into a glass or plastic vessel with lid. Check the pH of the solution. The pH of the
solution shall be between 7,0 and 8,0.
8.2 Chromatographic conditions
Determination of Cr(VI) is performed using the ion chromatographic technique. As the instrumental
equipment of the laboratories may vary, no specific applicable instructions can be provided for analysis.
However, the operating parameters and examples of the ion chromatographic analysis for Cr(VI) listed
in Annex A and Annex B have been successfully tested and used. Annex A determines Cr(VI) by direct
detection of chromate peak at 372 nm. Annex B determines Cr(VI) after a post-column reaction with 1,5-
diphenylcarbazide by measuring the absorption peak at 540 nm.
The method used should be verified using the recovery rate determination (8.4).
Record the injection volume as V and record the area of the chromate peak as A.
M
8.3 Calibration
The content of Cr(VI) in the sample of soil improvers and growing media is determined with an external
standard calibration.
The operator shall prepare calibration solutions from the standard solution (5.5). The Cr(VI)
concentration in these solutions should cover the expected range of measurement.
The following tasks shall be performed:
oSIST prEN 18257:2025
prEN 18257:2025 (E)
— Plot a suitable calibration curve by using at least five standards. Pipette the given volumes of standard
solution (5.5) into volumetric flasks. Make up to volume with the extraction solution or its dilution
(5.1), mix well, filter (6.10) and transfer a suitable aliquot volume into a vial (6.11).
— Transfer an aliquot to a suitable vial (6.11) corresponding for the chromatographic system (6.8).
— Inject the standards in the chromatographic system (6.8). Introduce the same volume for each
standard. It is recommended to inject equal volume for samples. Record the volume injected as V in
c
μl.
— Plot the Cr(VI) concentrations in micrograms of chromium per millilitre (μg/ml) against the
measured areas of the peaks of chromate. Plot the Cr(VI) concentration on the x-axis and the area on
the y-axis.
8.4 Determination of the recovery rate
Verify the method by preparing a soluble Cr(VI) standard solution from a stock standard solution from a
different source than that used for preparing the calibration
