SIST EN ISO 10519:2026 is the Slovenian adoption of International Organization for Standardization (ISO) 10519:2026, and it specifies a spectrometric method for determining the chlorophyll content of rapeseed. It is intended for laboratories and other users who need a reproducible way to measure chlorophyll in rapeseed samples; it does not apply to chlorophyll in oils.

What does SIST EN ISO 10519:2026 specify?

SIST EN ISO 10519:2026 covers one analytical method for rapeseed and is organized into 12 clauses plus two informative annexes. The method is built around sample preparation, solvent extraction, spectrometric measurement, calculation of chlorophyll content, and a reportable test result.

AnnexWhat it covers
Annex AResults of an interlaboratory test used to summarize precision data
Annex BPreparation of the calibration curve used to determine the k factor

The scope is narrow and practical: it is for rapeseed testing, not for oil analysis. That matters because sample type and matrix affect extraction and the way absorbance is interpreted.

What are the key requirements of SIST EN ISO 10519:2026?

SIST EN ISO 10519:2026 sets out a workflow that starts with representative sampling, then moves through grinding, extraction, measurement, and calculation. The main purpose is to make chlorophyll results comparable between laboratories.

Sampling and sample preparation

Clause 7 says sampling is outside the method itself, and refers users to ISO 542 for a recommended sampling method. Clause 8 requires preparation of the test sample in accordance with ISO 664, after impurities are removed.

For wetter seed, Clause 8 requires drying when moisture is above a mass fraction of 10 % before grinding. This reduces the risk of destroying chlorophyll pigments, which matters if the sample has been stored or transported in damp conditions.

Extraction and measurement

Clause 9 uses a 2 g test portion and 30 ml of extraction solvent. The extract is made with anhydrous ethanol combined with iso-octane, n-heptane, or petroleum ether, then shaken either for 1 hour in a tube with steel balls or for 20 minutes in a commercial ball mill.

The extract is allowed to settle, filtered, and measured in a spectrometer at 665 nm, 705 nm, and 625 nm. The extra wavelengths are used for baseline correction, so the result is based on corrected absorbance rather than a single raw reading.

Calculation and calibration

Clause 10 expresses chlorophyll content in milligrams per kilogram of product as received. The calculation uses corrected absorbance, optical path length, sample mass, solvent volume, and the constant k.

Clause 10 also states that k is 13 under the specified conditions, but if the spectrophotometer bandwidth varies, k may need to be determined for the chosen instrument and fixed setting. Annex B shows how to prepare the calibration curve for that purpose. In practice, this means users must know their instrument’s bandwidth and verify the calibration if needed.

Precision and reporting

Clause 11 gives precision guidance from an interlaboratory study: repeatability and reproducibility are stated as relative limits for comparing results. That helps laboratories judge whether two results are close enough to be considered consistent.

Clause 12 requires the test report to identify the sampling method if known, the method used, the result or results, the final quoted result if repeatability was checked, the test date, and any operating details or incidents that could have affected the outcome. This makes the report traceable and usable in quality records.

What terms does SIST EN ISO 10519:2026 define?

SIST EN ISO 10519:2026 defines a small set of technical terms for the method.

  • Chlorophyll content - the mass fraction of substances in the sample that absorb near 665 nm, expressed as chlorophyll A in milligrams per kilogram.
  • Corrected absorbance - the absorbance value used in the final calculation after baseline correction from the reference wavelengths.
  • Test portion - the weighed portion of prepared sample used for extraction and measurement.
  • Repeatability - the closeness of results from the same laboratory, operator, equipment, and short time interval.
  • Reproducibility - the closeness of results from different laboratories using the same method on the same material.
  • Bandwidth - the spectrometer characteristic that affects measurement resolution and the k factor used in the calculation.

Who uses SIST EN ISO 10519:2026?

SIST EN ISO 10519:2026 is used by analytical laboratories, oilseed testing labs, seed processors, quality managers, and buyers who need to compare rapeseed lots on a chlorophyll basis. It is also relevant to organizations that control raw material quality before further processing, because chlorophyll content is a measurable quality attribute in rapeseed.

The method is suited to routine laboratory work where the same procedure must be applied consistently across samples and across sites. Its precision section and test report requirements are especially useful for quality control, disputes, and specification checking.

What changed in SIST EN ISO 10519:2026 from the previous edition?

SIST EN ISO 10519:2026 cancels and replaces ISO 10519:2015. The document states that it is a minor revision and that it also incorporates ISO 10519:2015/Amd 1:2025.

The change listed in the foreword is the addition of Annex B, which gives the preparation of the calibration curve to determine the k factor.

Which standards are used with SIST EN ISO 10519:2026?

SIST EN ISO 10519:2026 refers to several companion standards that support sampling, sample reduction, and moisture determination.

  • ISO 648 - specifies the pipettes used for accurate 30 ml dispensing.
  • ISO 664 - gives the laboratory-sample-to-test-sample reduction step used in preparation.
  • ISO 665 - is used when chlorophyll content needs to be expressed relative to dry product.
  • ISO 542 - is cited as a recommended sampling method.
  • ISO 5725:1986 - underpins the interlaboratory precision study summarized in Annex A.

What does the SIST EN ISO 10519:2026 document contain?

SIST EN ISO 10519:2026 contains the full analytical sequence: scope, references, term definition, principle, reagents, apparatus, sampling, sample preparation, procedure, calculation, precision, and reporting. It also includes an interlaboratory precision summary in Annex A and a calibration-curve procedure in Annex B.

The document provides a practical laboratory method with specified solvent composition, equipment requirements, wavelengths, extraction times, and reporting items. For users, the useful parts are the procedure, the calculation approach, the precision information, and the calibration guidance for instruments with variable bandwidth.
