ISO/FDIS 25490 - Traditional Chinese medicine — Citrus reticulata pericarp
REDLINE ISO/FDIS 25490 - Traditional Chinese medicine — Citrus reticulata pericarp
ISO 25490 is an ISO standard for Traditional Chinese medicine that sets minimum requirements and test methods for Citrus reticulata pericarp, the dried pericarp of ripe Citrus reticulata fruit and related cultivars. It is intended for raw herbal materials in international trade, including Chinese materia medica and decoction pieces derived from this plant.
What does ISO 25490 specify?
ISO 25490 specifies how to identify, test, package, store, transport, and label Citrus reticulata pericarp used as a herbal raw material. It focuses on material quality and authenticity, so a buyer or laboratory can check that a lot is genuine, clean, and suitable for trade.
The document is organized into clauses for scope, normative references, terms and definitions, descriptions, requirements and recommendations, sampling, test methods, test report, packaging, storage and transportation, and marking and labelling. It also includes three informative annexes that support identification, assay, and harmonized national or regional requirements.
| Annex | What it covers |
|---|---|
| Annex A | Thin-layer chromatogram (TLC) identification of Citrus reticulata pericarp |
| Annex B | Determination of hesperidin content |
| Annex C | Reference information of national and regional requirements |
What are the key requirements of ISO 25490?
ISO 25490 combines visual, microscopic, chromatographic, and chemical checks. In practice, this means the material must look like Citrus reticulata pericarp, match reference identification tests, and meet specified quality limits before it is accepted.
Identity and appearance
Clause 5 starts with general characteristics: the material shall be clean, free from leaves and foreign matter, free from living insects, mouldy material, visible contaminants, rancid odour, and abnormal taste. These checks matter because they screen out damaged or contaminated lots before further testing.
Clause 5.2 describes the expected morphology, including the peeled lobes or slices, thickness, colour of the outer and inner surfaces, texture, odour, and taste. Clause 5.3 then describes the microscopic features of the powder and cross-section, such as mesocarp cells, epidermal cells, stomata, calcium oxalate crystals, hesperidin crystals, and vessels. These details help laboratories confirm identity and spot substitution or adulteration.
Chemical and quality criteria
Clause 5.4 uses TLC identification to compare the sample spot with the hesperidin reference spot. This gives a practical identity check that is especially useful for incoming inspection and disputes about authenticity.
Clause 5.5 requires foreign matter to be determined, and Clause 5.6 sets a moisture limit of not more than 13%. Moisture control matters because excess water can support mould growth and shorten storage life.
Clause 5.7 requires determination of hesperidin content, which is the main assay marker in this standard. Clause 5.8 requires determination of heavy metals such as arsenic, mercury, lead, and cadmium, Clause 5.9 requires pesticide residues to be determined, and Clause 5.10 requires aflatoxins to be determined, including aflatoxin B1, B2, G1, and G2. These tests address common trade risks - contamination from cultivation, storage, and processing.
Sampling, test methods, and reporting
Clause 6 directs sampling to ISO 23723:2021, Clause 8. Clause 7 then points to the test methods to use for each quality attribute, including macroscopic identification, microscopic examination, TLC identification, foreign matter, moisture, hesperidin, heavy metals, pesticide residues, aflatoxins, and sulfur dioxide. For a buyer or lab, this means the standard does not leave method choice open - it points to named procedures.
Clause 8 requires the test report to identify the sample, state the sampling method and test method used, give the results, record any optional operating details or incidents that may have affected the result, note any anomalies, and give the test date. This makes results traceable in audits, quality agreements, and shipment release.
Packaging, storage, and labelling
Clause 9 applies the storage and transportation requirements of ISO 22217. Clause 10 refers to ISO 21371 for labelling and requires packages to show the product name and Latin scientific name, the quality features determined under Clause 5, gross and net weight, country and province/state of origin, production date, batch number, expiry date, and storage and transportation methods. This is the information typically needed for receiving checks and traceability.
What terms does ISO 25490 define?
- Citrus reticulata pericarp - The dried pericarp of the ripe fruit of Citrus reticulata Blanco or the listed related cultivars.
- Chinese materia medica - Whole medicinal materials used as traditional herbal products in trade and practice.
- Decoction pieces - Prepared pieces derived from the plant for later use, especially in traditional medicine supply chains.
- TLC - Thin-layer chromatography, used here as an identification method by comparing sample spots with a reference.
- Hesperidin - The marker compound used for identification and assay in this standard.
- Foreign matter - Unwanted material mixed with the herbal raw material and measured as part of quality control.
- Aflatoxins - A group of fungal toxins that the standard requires to be determined because they affect safety.
Who uses ISO 25490?
ISO 25490 is used by producers, processors, traders, importers, exporters, laboratories, and quality managers handling Citrus reticulata pericarp. It is also useful for inspectors and regulatory bodies that need a common reference for authentication, sampling, labelling, and lot release.
In practice, suppliers use it to prepare and label material consistently, laboratories use it for identification and assay, and buyers use it to set acceptance criteria in contracts for Chinese materia medica and decoction pieces.
Which standards are used with ISO 25490?
ISO 25490 relies on several companion standards for the parts it does not define itself:
- ISO 23723:2021 - Provides general requirements for herbal raw material and materia medica, and Clause 8 supplies the sampling method referenced in ISO 25490.
- ISO/TS 21310 - Provides the microscopic examination method used for Clause 7.2.
- ISO 18664 - Provides the method for determination of heavy metals in herbal medicines.
- ISO 22258 - Provides the gas chromatography method for pesticide residues in natural products.
- ISO 22283 - Provides the LC-FLD method for aflatoxins in natural products.
- ISO 22590 - Provides the titration method for sulfur dioxide in natural products.
- ISO 22217 - Provides storage requirements for raw materials and decoction pieces.
- ISO 21371 - Provides labelling requirements for products intended for oral or topical use.
What does the ISO 25490 document contain?
ISO 25490 includes a botanical description of Citrus reticulata pericarp, with figures showing the plant structure and the microscopic cross-section used for identification. It also contains an informative TLC procedure in Annex A and an informative HPLC procedure for hesperidin in Annex B.
Annex C gives a comparative table of national and regional requirements, which is useful when aligning local specifications with the ISO standard. The main body also lays out the reporting, packaging, storage, transportation, and labelling expectations that support routine quality control and trade documentation.

Introduction
Citrus reticulata pericarp is also called Citri Reticulatae pericarpium, dried Tangerine Peel, Chen Pi in
Mandarin, or 陈皮 or 陳皮 (aged dried tangerine peel) in Han characters, which is the dried pericarp of the
ripe fruit of C. reticulata Blanco or its cultivars (Fam. Rutaceae), C. unshiu Markovich, C. reticulata 'Chachi', C.
reticulata 'Dahongpao', C. reticulata 'Unshiu', C. reticulata 'Tangerina'.
C. reticulata pericarp has been commonly consumed as a health food supplement and in traditional Chinese
medicine (TCM) for thousands of years due to its high nutritional and medicinal value. According to Shennong
Bencao Jing, written between 300 BC and 200 AD, one of the earliest books of Chinese medicine, C. reticulata
pericarp was regarded as one of the top-grade medicinal herbs that could extend one's life expectancy to
treat the symptoms of chest tightness and shortness of breath caused by reversed flow of internal heat in
the chest, and help with digestion and excretion. It can also remove bad breath, regulate qi function, and
make people feel energetic. Along with a growing number of studies on C. reticulata pericarp, it has been
utilized as a versatile medicinal herb with wide benefits in treating nausea, vomiting, indigestion, diarrhea,
cough, expectoration, preventing cardiovascular diseases, etc. Modern pharmacological studies indicate
that C. reticulata pericarp possesses wide therapeutical effects such as antioxidant, anti-inflammatory, anti-
cancer, and protective effects on the liver and nerves. Approximately 140 chemical compounds have been
isolated and identified in C. reticulata pericarp, including vitamins, minerals, polysaccharides, flavonoids,
volatile oil, alkaloids, phenolic acids, and limonoids.
[1]
Among 1607 formulations in the 2020 edition of the Chinese Pharmacopoeia , there are 180 formulations
containing C. reticulata pericarp (occupying 11,20%); among 294 formulations in the approval Standards
for OTC Kampo products from the ministry of health, labor, and welfare (MHLW) of Japan, there are 49
formulations containing C. reticulata pericarp (occupying 16,67%); among 2825 herbal preparations in
South Korea, there are 346 herbal preparations containing C. reticulata pericarp (occupying 12,25%).
Moreover, in the United States and Europe, C. reticulata pericarp is also a dietary supplement ingredient.
In China, Korea, and Japan, C. reticulata pericarp is also often used in food and beverages such as processed
into dried tangerine pericarp, preserved tangerine pericarp, tangerine pericarp tea, tangerine pericarp
wine, tangerine pericarp cakes, tangerine pericarp essential oils, tangerine pericarp condiments, etc.
Based on the statistics of the general administration of customs of the People’s Republic of China, the import
and export trade volume of C. reticulata pericarp ranged from 2 405 tons to 3 463 tons globally from 2018
to 2022, respectively, which was worth a total of 41 million USD. There are more than 30 trading countries
and regions of C. reticulata pericarp in the world, and the number is still increasing. From 2018 to 2022, the
top 5 countries with the largest import trade volume of C. reticulata pericarp were Vietnam, Spain, Japan,
Argentina, and Thailand.
Although C. reticulata pericarp is widely utilized and cultivated in a lot of countries and regions, especially
in China, Korea, and Japan, however, the quality standards of C. reticulata pericarp provided from different
areas or by different cultivators are quite different. In fact, C. reticulata pericarp has been recorded in the
[1] th [2]
2020 edition of the Chinese Pharmacopoeia , the Japanese Pharmacopoeia (18 edition) , the Korean
th [3] [4]
Pharmacopoeia (12 edition) , the United States Pharmacopeia (NF2023 edition) , and the European
th [5]
Pharmacopoeia (11 edition) , but these standards are not harmonized, limiting the international trade
of C. reticulata pericarp. Thus, there is a clear and urgent need to develop an international standard for
harmonizing the existing standards and ensuring the safety and quality of C. reticulata pericarp in the
international market.
As national implementation may differ, National Standards Bodies are invited to modify their national
standards' values in 5.6. Examples of national and regional values are given in Annex C.

China, Korea, and Japan, however, the quality standards of C. reticulata pericarp provided from different areas
or by different cultivators are quite different. In fact, C. reticulata pericarp has been recorded in the 2020
[1] th [2]
edition of the Chinese Pharmacopoeia ,, the Japanese Pharmacopoeia (18 edition) , the Korean
th [3] [4]
Pharmacopoeia (12 edition) ,, the United States Pharmacopeia (NF2023 edition) ,, and the European
th [5]
Pharmacopoeia (11 edition) ,, but these standards are not harmonized, limiting the international trade of
C. reticulata pericarp. Thus, there is a clear and urgent need to develop an international standard for
harmonizing the existing standards and ensuring the safety and quality of C. reticulata pericarp in the
international market.
As national implementation may differ, National Standards Bodies are invited to modify their national
standards' values in 5.6.5.6. Examples of national and regional values are given in Annex C.Annex C.
