ISO/FDIS 25585 is an ISO standard published by ISO for Notopterygium incisum and Notopterygium franchetii rhizome and root, called Notopterygium rhizome and root. It specifies the quality and safety requirements for this natural medicine in international trade, including Chinese materia medica and decoction pieces made from it. The document is written for users who need to identify the material, test its quality, and handle it consistently in trade and laboratory work.

What does ISO/FDIS 25585 specify?

ISO/FDIS 25585 specifies the quality and safety requirements for dried Notopterygium rhizome and root used as a natural medicine. It applies to material sold in international trade and excludes wild source herbs.

The document is organized into 10 clauses and 5 informative annexes. The main clauses cover scope, normative references, terms and definitions, descriptions, quality requirements, sampling, test methods, test reports, packaging and storage, and marking and labelling.

AnnexWhat it covers
Annex AIdentification of thin-layer chromatogram
Annex BDetermination of characteristic fingerprint
Annex CDetermination of total content of notopterol and isoimperatorin
Annex DReference of national and regional requirements
Annex EGrading information for Notopterygium rhizome and root

What are the key requirements of ISO/FDIS 25585?

ISO/FDIS 25585 combines visual identification, instrumental identification, chemical quality limits, and handling rules. In practice, it gives traders and laboratories a common basis for accepting, testing, grading, and labelling the herb.

Identity and appearance

  • Clause 5.1 requires the sample to be clean, free from rootlets, and free from visible living insects, mouldy material, and external contaminants. This matters because basic cleanliness is checked before deeper testing starts.
  • Clauses 5.2 and 5.3 describe the macroscopic and microscopic characteristics of Notopterygium incisum and Notopterygium franchetii. These descriptions help users distinguish the correct plant material from look-alikes and from mixed or damaged lots.
  • Clause 5.2 also distinguishes the named forms used in trade, including silkworm-form, nodular-form, bamboo-form, and strip-form material. That helps buyers and inspectors classify commercial lots by appearance.
  • Clause 7.1 requires random sampling of not less than 500 g from each batch, examined by visual observation in daylight and smell. This supports representative checking, rather than judging a batch from a few pieces.

Identification and chemical quality

  • Clause 5.4 requires TLC identification to show spots and bands specific to Notopterygium rhizome and root. This gives a simple identity check for extract preparation and comparison.
  • Clause 5.5 requires the HPLC fingerprint to match the relative retention time range of the reference fingerprint. In practice, the chromatographic profile is used to confirm that the material fits the expected chemical pattern.
  • Clause 5.6 requires marker compounds such as notopterol and isoimperatorin to be determined, with their total content not less than 0.4% by mass fraction. The annexed assay shows how this is measured and how the two markers are compared.
  • Clause 5.7 requires essential oil to be determined, with a minimum content of 14 mL/kg. This is a practical quality indicator for lots intended for trade and grading.

Safety limits, sampling, and reporting

  • Clauses 5.9 to 5.16 set limits or determinations for total ash, acid-insoluble ash, moisture, foreign matter, heavy metals, pesticide residues, sulfur dioxide, and aflatoxins. These checks help identify adulteration, contamination, and poor storage or processing.
  • Clause 6 points sampling to ISO 23723. That means the sampling step is aligned with a separate general ISO method for herbal raw material and materia medica.
  • Clause 8 requires the test report to identify the sample, state the standard used, show the sampling and test methods, record results, note any optional operating details or incidents, list anomalies, and give the test date. This is the record that supports traceability and review.
  • Clauses 9 and 10 control packaging, storage, transport, and marking. Packaging must not transfer odour or flavour or create health risk, and labels must state the quality features from Clause 5, masses, origin, production date, batch number, expiry date, and storage and transport method.

What terms does ISO/FDIS 25585 define?

  • Notopterygium rhizome and root - The dried rhizome and root of Notopterygium incisum and/or Notopterygium franchetii after primary processing, harvested in autumn or early spring when the aerial part withers.
  • Primary processing - The pre-treatment step that turns harvested natural material into medicinal material, including removing rootlets and clay and drying in the sun or oven.
  • Silkworm-form Notopterygium rhizome - A rhizome subtype with a stout cylindrical shape, dense nodes, and a form resembling a silkworm.
  • Nodular-form Notopterygium rhizome - A rhizome subtype with irregular nodular swellings and multiple stem scars at the apex.
  • Bamboo-form Notopterygium rhizome - A rhizome subtype with elongated internodes and sparse nodes, resembling bamboo joints.
  • Strip-form Notopterygium rhizome and root - A rhizome-and-root subtype with an elongated strip-like form and a tapering root tip.
  • Qiang Huo - The Chinese Pinyin name for Notopterygium rhizome and root.

Who uses ISO/FDIS 25585?

ISO/FDIS 25585 is used by people who buy, process, test, or inspect Notopterygium rhizome and root in traditional medicine trade. Typical users include herbal-material producers, processors, quality-control laboratories, importers and exporters, and buyers who need a clear specification for acceptance.

It is also relevant to laboratory staff who run TLC, HPLC, ash, moisture, contaminant, pesticide, sulfur dioxide, and aflatoxin tests, and to organizations that write product specifications or grade the material for commercial supply.

Which standards are used with ISO/FDIS 25585?

  • ISO 23723 - Provides general requirements for herbal raw material and materia medica, and it supplies the sampling method plus several test methods used in this document.
  • ISO/TS 21310 - Provides the microscopic examination method used for microscopic identification.
  • ISO 18664 - Provides the heavy metals determination method.
  • ISO 22258 - Provides the pesticide residues determination method by gas chromatography.
  • ISO 22283 - Provides the aflatoxins determination method by LC-FLD.
  • ISO 22590 - Provides the sulfur dioxide determination method by titration.
  • ISO 21371 - Provides labelling requirements for products intended for oral or topical use.
  • ISO 16791 - Provides requirements for machine-readable coding of medicinal product package identifiers.
  • ISO 21300:2019 - Is cited in the terms section as the source for the definition of primary processing.

What does the ISO/FDIS 25585 document contain?

The document contains descriptive material for the plant and product forms, including a structure figure and microscopic schematic. It also contains practical test procedures for identity and assay work.

Annex A gives a TLC procedure and figure for identifying notopterol and isoimperatorin. Annex B gives the HPLC fingerprint procedure, including system conditions, chromatographic elution, and a reference fingerprint figure. Annex C gives the HPLC assay for total notopterol and isoimperatorin, including calculation formulas and system suitability. Annex D summarizes national and regional quality requirements, and Annex E gives grading information based on appearance and essential oil content.

The main body also specifies the test report content, packaging and transport protections, and marking and labelling items, so the standard can be used not only for testing but also for release and trade documentation.

Introduction
The medicinal history of Notopterygium incisum and Notopterygium franchetii rhizome and root (abbreviated
as Notopterygium rhizome and root, Chinese Pinyin name Qiang Huo) dates back to over 2 000 years.
Notopterygium rhizome and root is used as a traditional herbal medicine in China, Japan, South Korea, the
United States, Netherlands, and other countries. It has played a role in various therapeutic effects, such as
an anti-inflammation, analgesic, antipyretic, antibacterial, antiviral, anti-arrhythmia, and anti-thrombotic,
intended for the treatment of common cold, headache, rheumatic pain, and rheumatoid arthritis, etc.
Origin plants of Notopterygium rhizome and root are Notopterygium incisum K. C. Ting ex H. T. Chang and N.
franchetii H. de Boiss. (Fam. Apiaceae) as recorded in several pharmacopeias, such as Chinese Pharmacopoeia,
Japanese Pharmacopoeia, Korean Pharmacopoeia, and European Pharmacopoeia. In addition, the Korean
Pharmacopoeia contains Ostericum koreanum rhizome and root for local use.
The establishment of an international standard for Notopterygium rhizome and root is necessary to
guarantee the clinical effectiveness, patient safety, and controllability in global commerce and trade.
As national implementation may differ, national standards bodies are invited to modify the values given in
5.6, 5.7, 5.9, 5.10 and 5.11 in their national standards. Examples of national and regional values are given in
Annex D. In addition, grading information of Notopterygium rhizome and root are given in Annex E.
